چکیده

Tetanus neurotoxin (TeNT) is one of the most poisonous substances on Earth and a major cause of neonatal death in non-vaccinated areas (1). TeNT targets the neuromuscular junction (NMJ) with high affinity, yet the nature of the TeNT receptor complex at the NMJ remains unknown (2). Here we show that nidogens (also known as entactins (3)) are highly enriched at the basement membrane of the NMJ and are essential for TeNT binding to motor neurons, where TeNT enters the central nervous system to exert its toxic effects. Inhibition of the TeNT-nidogen interaction using small nidogen-derived peptides or genetic ablation of nidogen-2 protects mice from TeNT-induced spastic paralysis. Our report is the first to demonstrate the direct involvement of a protein receptor for TeNT at the NMJ, paving the way for the development of novel therapeutics for the prevention of tetanus targeting this novel protein-protein interaction.

P 115: Potential Therapeutic Targets Related to Neuroinflammation in Treatment and Prevention of Autism

Autism spectrum disorder (ASD) is a mental condition, present from early childhood, characterized by great difficulty in communicating and forming relationships with others and using language. In the last four decades many studies have shown that immune responses in different regions of brain play an important role in ASD pathogenicity. A conservative estimate based on the research suggests tha...

Background and Objectives: Tetanus is a lethal disease caused by Gram-positive Clostridium tetani bacteria toxin. The annual incidence of this disease in the world is one million people per year [1]. Typical symptoms include trismus, risus sardonicus, and opisthotonos. The main cause of death in the primary phase is laryngeal muscle spasm and in the late phase is dysrhythmia [1]. Cloning and expression of tetanus toxin C fragment (Fc) in prokaryotic vector for constructing recombinant protein based vaccine for tetanus

Tetanus is a disease caused by tetanus toxin, a potent inhibitor for the release of inhibitory neurotransmitter in the central nervous system that causes spastic paralysis. Fragment C (52 kD) of this toxin is responsible for binding to the neuronal membrane. For this reason, and also its non toxigenic and immunogenic nature, this fragment might be ideal for new vaccine development. Presently, w...
